I am available most of the times on skype and have to receive and make calls, but for that my system needs to run 24 hrs long. I mean is there any way out, that when my computer is off and I receive a call on skype, It wakes up automatically. Simply give me a setting to make skype wake computer I use.

 It seems that there is no way to wake computer from Hibernation or Sleep mode, yet. Skype did not implement this feature. For my part, I too hope for this feature. It would be very convenient and cool. You could be away from the computer and when the call or message comes in, Skype automatically wakes computer up. But the reality is that it's not possible to set it up like that.
Maybe in the near feature Skype will have this, so we can only hope for now.
