In computer networking, Server Message Block (SMB), also known as Common Internet File System operates as an application-layer network protocol mainly used for providing shared access to files, printers, serial ports, and miscellaneous communications between nodes on a network. But I do not understand that how it is working? Please anyone express me about this.

In networking term, Server Message Block SMB or also know as CIFS Common Internet File System, Yes it is an ApplicatIon Layer network Protocol, As you said above Correctly, SMB generally used on Microsoft Windows operating system.
